چکیده

Background: Osteonecrosis of the femoral head (ONFH) is an ischemic disease characterized by impairment angiogenesis. We have previously elucidated role tsRNAs and BMSC exosomes in ONFH, but whether tsRNA-modified promote angiogenesis ONFH remains unclear. Methods: The expression angiogenesis-related tsRNA plasma from patients was examined q-PCR. function HUVECs identified CCK-8 assay. Exosomes purified tsRNA-15797 overexpressed BMSCs were cocultured with to examine their molecule mechanism tsRNA-15797-modified explored RNA sequencing, dual-luciferase assay, immunofluorescence. Results: A tRNA-derived small down-regulated patients. found effects BMSCs-derived on accelerating migration, which further enhanced after overexpressing tsRNA-15797. Besides, overexpression would lead down-regulation LFNG correlated could directly interact LFNG. demonstrated that LNFG weakened pro angiogenic migratory exosomes. Conclusion: successfully constructed BMSC-derived it induced targeting Thus, tsRNA-15797-loaded may be a potential target therapy drug for ONFH.

Osteonecrosis of the femoral head.

New cases of osteonecrosis of the femoral head in the United States number between 10,000 and 20,000 per year. This disease usually affects patients in their late 30s and early 40s. Although a number of authors have related specific risk factors to this disease, its etiology, pathogenesis, and treatment remain a source of considerable controversy. This disorder has been associated with corticos...

Exosomes from Human Synovial-Derived Mesenchymal Stem Cells Prevent Glucocorticoid-Induced Osteonecrosis of the Femoral Head in the Rat

Osteonecrosis of the femoral head (ONFH) represents a debilitating complication following glucocorticoid (GC)-based therapy. Synovial-derived mesenchymal stem cells (SMSCs) can exert protective effect in the animal model of GC-induced ONFH by inducing cell proliferation and preventing cell apoptosis. Recent studies indicate the transplanted cells exert therapeutic effects primarily via a paracr...

Arthroplasty in Femoral Head Osteonecrosis

Osteonecrosis of the femoral head is a destructive joint disease requiring early hip arthroplasty. The polyethylene-metal design using a 22-mm femoral head component, introduced by Charnley in 1950, has been widely used for over half a century. Since then, different materials with the capacity to minimize friction between bearing surfaces and various cement or cementless insert fixations have b...

Available Findings Fail to Provide Strong Evidence of the Role of Bone Morphogenic Protein-2 in Femoral Head Osteonecrosis

Despite widespread research on non-traumatic femoral head osteonecrosis (FHON), there is no consensus about preventative treatment options. Insufficient blood supply and increased intra-osseous pressure are the initiating events in the majority of cases. BMPs are growth factors that belong to the transforming growth factor β (TGFβ) superfamily. Two specific formulations of BMPs have already bee...
